Ac-Tle-Tle-Asn(Me)₂-Ala-AMC is the shortest human cytomegalovirus (HCMV) protease substrate to date. It displayed excellent kinetic properties with kcat and Km values of 0.035 s⁻¹ and 13.2 µM, respectively. The resultant specificity constant kcat/Km of 2650 M⁻¹ s⁻¹ was 10-fold greater than for the non-improved substrate Abz-Val-Val-Asn-Ala-Ser-Ser-Arg-Leu-3-nitro-Tyr-Arg-OH.
