| 实验参考方法: | 3xHA标签蛋白纯化方案 A:细胞收集和裂解 1. 收集表达Sld7-3Flag-HA的YTT4细胞(7×10⁷cells/ml, 2.5L)。 2. 用蒸馏水清洗细胞一次。 3. 使用砂浆研磨机在液氮下破坏细胞。 4. 将细胞裂解液悬浮在5ml裂解缓冲液A(50mM HEPES-KOH(pH 7.5),300mM氯化钾,0.5%吐温20, 0.05% NP40, 10%甘油,1×Complete, 1%蛋白酶抑制剂混合物,50mM氟化钠,2mM β-甘油磷酸盐,0.4mM Na3VO4, 0.5mM Na4P2O7,1mM PMSF)中。 5. 在4°C下,12,000rpm离心15分钟,澄清裂解物。 B:蛋白提取和吸附 6. 将蛋白提取物吸附到1ml Sepharose 4B中,在4°C下吸附30分钟。 7. 将混合物与1ml anti-Flag M2亲和凝胶混合后与含有5mg/ml BSA的裂解缓冲液A预孵育,在4℃下孵育2.5小时。 C:用Flag Peptide洗涤和洗脱 8. 用含有5mg/ml BSA的ml裂解缓冲液A洗涤三次。 9. 用500μl的Flag洗脱缓冲液(裂解缓冲液A含有150mg/ml的3× Flag肽)在4℃下孵育1小时,洗脱结合蛋白2次。 D: Anti-HA Matrix进一步纯化 10. 洗脱液与120μl Anti-HA Matrix 混合后用anti-Flag M2亲和凝胶预处理,4℃孵育2.5小时。 11. 用1ml含5mg/ml BSA的缓冲液A洗涤3次,1ml含0.1mg/ml BSA的缓冲液A洗涤1次,1ml缓冲液A洗涤1次。 12. 用100μl HA洗脱缓冲液(裂解缓冲液A含1.5mg/ml 3× HA肽)在37℃下孵育30分钟,洗脱结合蛋白两次。 E:蛋白沉淀和分析 13. 加入三氯乙酸沉淀蛋白,然后离心。 14. 沉淀在50μl的1× SDS上样缓冲液中煮沸3分钟。 15. 用SDS -聚丙烯酰胺凝胶分离蛋白质,用SilverQuest染色。 16. 切胶获取蛋白,用胰蛋白酶消化,并用质谱法分析。 本方案仅供参考,请根据您的具体需要进行修改。 References: [1] Tanaka, T., Umemori, T., Endo, S., Muramatsu, S., Kanemaki, M., Kamimura, Y., Obuse, C., & Araki, H. 3X HA Tag is a bioactive peptide composed of the 9-amino-acid sequence YPYDVPDYA repeated 3 times[1]. HA Tag is derived from the hemagglutinin (HA) surface glycoprotein of human influenza virus, featuring high specificity and hydrophilicity. It enhances the possibility of antigen - antibody reactions on the surface of fusion proteins[2]. With more repeats, 3X HA Tag has stronger antigenicity and higher binding affinity for anti - HA antibodies, ensuring higher detection sensitivity, especially for low - abundance proteins[3]. It's widely used in protein affinity purification experients to elute HA-tagged proteins[4][5].

1、合成CTC树脂:称取2.03g CTC Resin(如初始取代度约为0.65mmol/g)和1.58mmol Fmoc-Glu(OtBu)-OH于反应器中,加入适量DCM溶解氨基酸(需要注意,此时CTC树脂体积会增大好几倍,避免DCM溶液过少),再加入3.96mmol DIPEA(Mw:129.1,d:0.740g/ml),反应2-3小时后,可不抽滤溶液,直接加入1ml的HPLC级甲醇,封端半小时。依次用DMF洗涤2次,甲醇洗涤1次,DCM洗涤一次,甲醇洗涤一次,DCM洗涤一次,DMF洗涤2次(这里使用甲醇和DCM交替洗涤,是为了更好地去除其他溶质,有利于后续反应)。得到 Fmoc-Glu(OtBu)-CTC Resin。结构图如下:

2、脱Fmoc:加3倍树脂体积的20%Pip/DMF溶液,鼓氮气30分钟,然后2倍树脂体积的DMF 洗涤5次。得到 H2N-Glu(OtBu)-CTC Resin 。(此步骤脱除Fmoc基团,茚三酮检测为蓝色,Pip为哌啶)。结构图如下:

3、缩合:取3.96mmol Fmoc-Leu-OH 氨基酸,加入到上述树脂里,加适当DMF溶解氨基酸,再依次加入7.92mmol DIPEA,3.76mmol HBTU。反应30分钟后,取小样洗涤,茚三酮检测为无色。用2倍树脂体积的DMF 洗涤3次树脂。(洗涤树脂,去掉残留溶剂,为下一步反应做准备)。得到Fmoc-Leu-Glu(OtBu)-CTC Resin。氨基酸:DIPEA:HBTU:树脂=3:6:2.85:1(摩尔比)。结构图如下:
